95% of the time, off... fully counter clockwise.

On rare occasions (either an extremely small, dry room, or using a VERY midrange voiced cab like a Schroeder), I will dial the VPF (contour/enhance) up to 8 or 9 o'clock, and then use the VLE at about the same level to eliminate the extra upper treble added by the VPF.

I start with everything flat & the filters off. Honestly, I play 90% like this. I'll be more likely to turn up the low pass filter depending on the specific tone I'm after, so it varies from moment to moment. I might bump the mid cut to smooth out any honkiness if I'm playing slap.
The EQ is great, but it's easier to grab one of the filter knobs on the fly. Besides, it really does sound best flat for my situation.

I really really like the bridge p/u; which tends to thin out my sound some, so I add a little bit of VLE and VPF to take the edge off and warm things up a tad.......but sparingly........
I like the tightness of the bridge, but like to add some beef to it with a touch of these knobs 7-10 0'clock...depending
if I were not leaning so hard on the bridge p/u, I would likely have these closer to off position.............
very useful knobs IME.....

VLE 9:00 to 10:00

VPF 8:00

I go for a p-bass with flats sort of tone with subdued highs.
