i generally use 40-100 dr hi beams. i am joining a new band where they go to drop D. any time my current band has done drop D it does feel a little floppy. will going from .100 to .105 going to make a huge difference? and will the thicker string die off quicker?

.100 to .105 will make a HUGE difference with the floppyness in drop d. I really don't think the thicker string will die off quicker, but it'll probably take a bit of time getting used to the slightly higher tension in standard tuning. If your just talking about replacing that one string, then it won't be a big change to get used to at all.

Buy yourself singles if you can. It'll help playability a lot - if you're using .40's now, buy yourself a set in gauges 40-55-70-105. That'll give you even tension all across!
