If just one string won't stay in tune the problem could be with that tuner. It might be slipping and need tightening or you could have way too many windings around the post or not enough and the string is slipping off (but that last would be rare).

Another possibility, with an old style Fender bridge in particular, is the height adjustment screws might be slipping and that saddle is unscrewing down with vibration. That used to be a problem with my early Fenders.

So, you have to find the source of the problem to answer your question. You can get a good setup job done for $40-$50, but if the tuner needs replacing, that would up the cost. A set up usually just includes setting the action, adjusting the truss rod and maybe the pickup height. Extra work costs more.

If you're completely new at this. take it to a good tech and get a free assessment and estimate.

^^^62bass is spot on.

Sometimes a false string can cause the problem. It does not matter if the string is brand new. It can still be false. If all else fails, change the string. Take it back to the retailer if it is a new one. They should be willing to give you another. It costs them nothing because the manufacturer will replace it to their inventory on their next order.
