Tune away, the truss rod adjustment and overall tuning will affect each other, so you will want to be "in tune" after you adjust the truss rod, and touch up after each adjustment. Most importantly, don't overdo it on the truss rod, a little goes a long way, and you need to go slow so that nothing breaks.

Assuming the truss rod is what actually needs adjusted (not some other aspect of your setup), you may be going the wrong direction. Go ahead and turn it back, retune, and recheck. A little better but not quite? Go another fraction of a turn and retune, recheck. You can do this all in one day, no need to wait a day.

Joshua, maybe you'd be willing to weigh in regarding my feelings about the wait time.

I always figure an hour or two is plenty for double rods or curved single rods. It's only compression rods that need 8 hours to see the response.

What do you think?

As far as wait time, I have some basses that a turn appears in seconds. Others take a little time, maybe not a day but sometime I will let it sit for a day before I screw with anything else.

See right there thats a great reason to own what some would call to many basses.

well since i adjusted the rod the 4th and 5th frets of only the E string have been constantly buzzing. Since then ive set it back, tightened it, loosened it, etc. each with saddle adjustments. After every adjustment, whether it is 1/2 turn or not even 1/8 turn i can adjust the string height for every string to not be buzzy except for the E string at the 4th/5th fret.

i adjusted it in the first place because my string height was a bit too high and just lowering the saddles made it buzz everywhere, i didnt even give it 1/2 of a turn that day and at first everything sounded ok, then the next day the buzzing started.

how much would it cost to get it set-up at a shop on average? I'm tired of trying to fix it lol.
