First if you are not already you need to listen to the Blues to understand the function of the bass in tat context. Next you have to learn the Blues progression. At it's most basic That's the I, IV and V chords of a given key played as follows: I I I I, IV IV I I, V IV I V. That's one measure for each chord symbol. Then learn some different lines based on the one, three, five, six and seven tones of each chord. Most blues bass lines repeat the same pattern over each chord at for a full chorus, When you can play the aforementioned comfortably and for five—ten minutes at a time then learn the Blues Scale and the Major and Minor Pentatonics.

As I said this is a very basic description and there are hundreds of variations, but if you learn the above and listen to a lot of blues, the variations will become apparent.

Need to know a minor pentatonic, major pentatonic, major and minor scales. Also, knowing the mixolydian mode (major scale with a flat seventh) helps, too.
In blues, the bass is the timekeeper so improvising is limited but if you listen to Tommy Shannon, he could do the timekeeping and still come up with some tasty lines so grab some SRV CD's and start listening.
Also, learn how to shuffle...very essential for blues and it used in a lot of other styles besides the blues.
