Considering that it sold a gajillion copies, you might call it the "most popular SONG with a distinctive bass line".

I would have guessed that it's a synth playing the bass line, but according to this article it's Louis Johnson on a Yamaha bass.

Oh no, no, no. It is very memorable, but in no way is it the most memorable or the most popular.

And I cite the Poll/Thread started by our friend Burk48237 as evidence of why I feel this way;

BEST "POP" BASS LINES

The minute you said, "Billy Jean" and "bassline", I instantly could hear the bassline in my head ( ||:bim bum bum bum bim bum bum bum :|| ) - so it is definitely a very memorable line - however, as Burk's thread demonstrates, there are SO MANY other lines that shine, your statement,

I'm listening to it right now, and I still suspect it's a synth. Or at the very least, there's a synth playing along with a real bass. It's just so rigid and MIDI-like in its tempo, and I hear that synth-y tone.

On the other hand, there's definitely a human-played bass on the bridge (the "People always told me, be careful what you do...." sections).

Well, if you judge by what 14 year olds try to play in music stores, it's either the RHCP version of Higher Ground, or Pink Floyd's Money.
Money REALLY amazes me sometimes. It's NOT hard, but I've heard alot more wrong versions than right!
