Having worked in various biology labs and done many DNA extraction and PCR experiments I can honestly say that if you know what you're doing you can isolate DNA, perform PCR, load it, run electrophoresis, and get results in 6 hours.

This is not for human samples nor is it for precise matching of an individual. This is for a standardized, quantitive, multi pass PCR used to identify levels of 36 different species of fungal mold. The test is performed to a high standard and the chain of custody, record keeping etc must be to a level that will stand up in court.
Last time I used them, I spoke to the tech in the mid to late morning and he said he had just got my samples and was preparing them. I had the results in my email by the end of business that day.

I had the results in my email by the end of business that day. | Admittedly I worked almost exclusively with Thalia dealbata but even still it was a 6-hour turnaround when using a kit. I assumed DNA isolation kits for blood, which I'd imagine are used much more frequently, would work just as quickly. |

The old Taq enzyme would be suspect, but advances have made this accurate. Thermal cyclers can be run in less than half a day the restriction enzyme incubation in less than two hours. Gel run for specific markers in about 30 min. Then fragments will need to be run for GATC accuracy - depending on the equipment... About two hours. To ensure accuracy you would want at least three labs running this simultaneously to eliminate any human errors (pipetting, etc). I suspect the military had their own and probably contracted verification with labs close by (India, China, etc). Pretty basic procedure.
