There was a discussion on the MIMF about the actually material used by Fender for the dots. The general consensus is that it was an undyed fiberboard similar to the material used in their pickup bobbins. The tan coloring comes from the rosewood leaching oils into the fibers.

I’ve had good luck using MDF for substitute clay dots. I’ve also heard of using the tan PVC from the plumbing supply

You could also use any one of a number of polymer clays like Sculpey to make the dots.
