Okay, so I have my pedal board starting with:
Wireless > Tuner > Fuzzrocious Rat King > Fuzzrocious Broke Dick Peanut Gallery > ISP
And I have a Sansamp RBI and SVT 4 Pro going into a matching 810E.

If I wanted to split the signal into a distorted dirty channel on the top cab coming from just the dirt pedals on my board and the RBI and a straight clean signal on the bottom cab from just the 4 Pro would all I need is this?

    I'm playing Spector basses, btw.

And of course I'd want to be able to mute both cabs when I step on my tuner.

So it'd be Wireless > Tuner > Radial (Output A) > Rat King > BDPG > ISP > RBI > Power Amp A input of 4 Pro > top cab

And Radial (Output B) > 4 Pro front input > bottom cab from power amp B

Would this ruin anything or is there something I should be aware of?

Yes that's all you need. Thread'd be better in the FX forum.
