my E string does not tune up, no parts are broken or loose but its permanently in drop A

I think that's your problem, then. The string is slipping.

When you tune up, does the pitch go up and then begins to go down again?

If yes, the problem is the string is slipping. In my opinion, of course.

I'm not sure, of course. At some point it should come off of the tuning peg, if that's was the case.

 

That might be a problem of. The tuning gear then.

2 full turns at least on Fender style basses. Also make sure you insert the end of the string into the center hole.
There is a part missing on the top of this tuner.

 

You have more problems than a slipping string.
Post more pics of the complete headstock.

Edit:If the back plate in the above pic is from your bass,you are missing some mooring screws.

 

Cheap 'Fender' style 4 on a side tuners are often not built to close enough tolerances, and can suddenly skip a few cogs as the string gets tighter. I quick fix for them is to take it apart, and reassemble without the leaf spring under the gear. This pulls the gear down enough to mesh with the worm gear a little better. It's a temporary measure til you can replace them.

 

You need tuner ferrules, there should be no space around the peg. May not solve the entire problem, but definitely a problem.

 

RyanOh has it. The ferule is missing. The post is bending under the tension from the string allowing th cogs to slip.
