Do I have a junk bridge?

It sounds like the pressure from the string is holding your saddle down tight, and that you're just backing the screw out without moving the saddle...is this what you meant? If so, try turning the screw a tad, then moving the saddle with your fingers...or, downtune a bit, let the saddle move, retune, then check intonation.
