Testing an Amphead with Combo unit?

Hey Guys,
I just received my GB Shuttle 6.2 and I want to do a test to make sure it works. I don't have a Cab yet, as I am going to build one, but I do have a JBL 18" powered Sub, an Fender Twin Reverb Tube Amp (80W)with 2 12" 4ohm Fender speakers and a little Marshall Combo tube amp (30W) with a single 12" Celestion Speaker.
Is there a way that I can plug my amp into any one of these units and just test for sound to make sure all is well with my head. It may be about 2-3months until I get my Cab built.
If so, do I power the amp up or not? Can I use an instrument cable for the test or not? I'm a complete idiot in this new world that I have entered into and I just don't want to damage anything. Please know that I do not intend to play with this test setup......again, just want to make sure my head works OK. I bought the head online from an out of state dealer. Thanks Guys
