Would playing a song that has a drop D tuning on my bass which is tuned standard sound totally different? I don't know exactly how to tune down D or anything.

meh you would definetly notice it if you were playing it alongside the recording. If you are practicing at home it shouldn't matter.
Only problem is that you won't get to the low D, but apart from that the notes are the notes.
To tune to Drop D, tune your E string to D. All other string remain the same.
