Sure it would. Nor is dust much of an enemy of these jacks unless it's really dusty.
No, really, it wouldn't. I know the circumstances of my use of the amp. Dust would get in while I'm using it. I'm not going to spend money on a custom made cover and keep the cover on while I play. Thanks for the suggestion but no thanks, it will not work.

I've thought about how to protect the amp and the solution is to get plugs. That will work. So if you have something on topic you can suggest, please go ahead.

I just ordered a pack of these off amazon recently. A pack of 25, only $7. I got them for my boss loop pedal since I only ever use it for bass, but it has 3 inputs on the right side and I get confused between them.

Do you know anyone with a 3d printer? It would be really easy to produce those.

Can you squirt RTV sealant into the unused ports or do you anticipate needing them in the future?

Rubber plugs fine, RTV waaaaay too permanent of an idea.
For any plug I would not go any longer than about 3/8 of an inch.
Any longer and you could be hitting contacts and opening or closing contacts
on some jacks.

If this is really an important thing, check out the rubber stoppers (like corks) in the nut and bolt aisle at any decent hardware store. Personally, I don't worry about it, but it's not my amp in question.

Hillman is a common supplier to hardware stores and home centers, I've used their stoppers to seal exhausts and intakes in r/c engines in storage.

never use standard RTV on electronics, the acetic acid can damage the contact plating. There are specialty products but not commonly available.

Maybe the next gig for the OP is Burning Man? :laugh:

If that’s true, nothing is going to help. Same for rodeo events, those who have worked these event know exactly what I mean,
