Fret at the 12th fret and compare it to the 12th fret harmonic.

I’ve always played a harmonic at the 12th fret, and tuned that to the desired pitch; then I’ve fretted a note normally at the 12th fret, and adjusted the intonation until the fretted note played in tune.

That seems to be a hold over from anchient guitarist ritual. I have never seen the need to do this on bass. Just press it how you normally would has always worked for me.

Yes fret it as you usually would, compare it to your 12th fret harmonic and adjust until they match. It doesn't make a huge difference but is "the right way to do it." It’s also best if you set the intonation in the same position you're going to be playing it but unless you use a hootenanny strap button it doesn't matter that much.

I guess that works if not using a Virtual Strobe tuner. I find with the Peterson Strobo Stomp’s 1/10 of 1 cent accuracy it is about as precise as one can get
