Does putting a cab on its side make a difference on the sound?

Maybe depending on the port location. It could acoustically throw the tones around the room differently.

It doesn’t matters.

Radiation from a port is omni-directional, so front or rear it's equally affected by cab placement. The only exception is if a rear ported cab is pushed tight to a wall. Omni-directional port output is why a port can be placed anywhere except directly behind the cone, as that will allow midrange to go through the port and that can cause problems with reflected midrange off a rear wall.

See the answer on another topic by one of the speaker guru’s. About front or rear ported cabs. Unless, see the exceptions. The room (or outside) however can have a huge impact on how it sounds.

The short answer is if it is front ported, yes. If there are multiple speakers, having them vertical as oppose to horizontal may help you hear it better. There might be a slight noticeable effect of having a tweeter or horn on the bottom or top depending on how far away from the cab you are. That said, once the band starts playing, you might not notice much difference unless you are playing too loud in relation to the rest of the players.

As for what the audience hears, I would find it extremely doubtful it would make any difference at all to anybody 10ft or further away.
