Probably 15 to 20 seconds to tune down the E string to Eb. But if the question is if a transposition down a 1/2 step the question then becomes, is an Eb required? If not, time required to retune is 0 seconds. If it is, then I'd just tune down the E to a D (ten seconds, used to doing it) and access the Eb that way.

I don't downtune. I bought a 5-string for times I need something on the B string. Get's to be a real pain to down tune for one song, up-tune for the next, and down tune for a 3rd song again. Saved my gig money and bought a 5-string specifically for those songs. Amazing what $219 could buy in a 5-string back in 2009. Will never replace my '82 Jazz, but I don't even have to make any adjustments (such as gain) on my amp with my 5-string in passive mode. I also use a wireless so I just unplug one and plug the other in.

The first comment explains it’s E to Eb. Your way means jumping up 11 rather than down 1.
