To lock or not to lock... that is the question.

But, as a rule, when you're building a bass line in a band situation, where do you like to begin? Or does it even matter?

Depends on the song, the style, and the goal.
When I have a bassline with a strong melody the drummer has to lock in with me! When the drummer plays an awesome groove, I always try to give that groove a melodie.
