My first thoughts are that we saw this kind of weight loss before in mice with fatty acid synthase inhibitors, which, if I remember correctly (it was more than 20 years ago) eventually were shown to be acting through toxicity. On glancing through the figures, it seems as if the authors did a pretty good job controlling for that. The upregulation of PPAR pathway (alpha, beta/delta, but not gamma) is intriguing, but the difference between mice and human in PPAR effects can be very murky (cardiac liabilities).
