Tuner (I like to look at the lights), compressor, and pre-amp. That's all I have on my board (besides an A/B/Y switch, which isn't an effect). I keep telling myself I'm going to get an EQ, but just haven't gotten around to it. I guess if it were really important I would have taken care of it already.
EBS Multi Comp, HPF/LPF and a Southampton All Things Equal. This is an always on buffer with two volume levels plus it has the OD from the Fifth Gear built in. Turning it on Dirty adds a wonderful gain stage that makes my bass come alive. There is an internal gain control that I have never opened up to fool with but however it came factory set really makes my sound punchy. I was using a Darkglass Vintage Microtubes as an always on OD with the Drive and Blend set pretty low but the Southampton really does what i wanted. I originally wanted it to have two volume levels for either solos or when using my envelope filter but the gain stage is a huge added feature.
For me, it's all about preamps. My always always on's are DOD fx10, Rusty Box, B7KU as well as an FDeck HPF under my board. I run dirt a lot so my DG X7 or Damnation MBD is on like 80 to 90% of the time too. The preamps are always on for tone shaping and coating my whole signal and anything that is in it with goodness. It also boosts my signal a little too. My amp is an all tube head that likes all the added color.
I do actually keep a TC spectracomp on all the time too. Thats for comp and a little boost and high end sheen.
Lately it's been (everything that is on always) Line6 G70 to BJFE Blueberry OD to Rodenberg GAS707B Boost to Goodwood Bass Interfacer to SFX Micro Thumpinator to amp.
Markbass Compressore. Always. Tube compression, levels out spikes, improves sustain, adds warmth.
(Snark Tuner is always plugged into tuner port and used every time I play. I usually just leave it on, as it has no effect on signal and I have it on a power supply, so no battery worries. But yeah, being in tune...)

I always have at least one dirt pedal on. It was primarily the VMT but that recently switched to the one control green bassmachine. Next up is the MBD-1 an finally the Gasoline. I will stack them too.
I use a Boss TU-3 Tuner, Keeley Bassist Compressor (always on, set and forget) and a VT bass deluxe (always on but switch between six programmed settings, the first setting is used about 80% of the time.)
Subharmonic Synth, EQ and compression are always on. Does it matter that they are rack mounts and not pedals?
My two always on effects for my band are the SpectraDrive for preamp/comp and Microtubes X for clank and grind. I'll swap to OD for mellower songs.
I rarely play anything clean these days, but when I do, its often in a jazz setting and I'll still play through the SpectraDrive.

i have no "always on" rules/effects, but a compressor can be a useful tool, most of the time.
