Dialing in like an idiot.

I say start out with the amp flat and have someone else you trust and is familiar with your band adjust it while the band is playing. Use a camera or cell phone to take pics of various settings so you can get back to them as things get tweaked. The advice above about using either pickup by itself is great advice. I almost never, except when slapping, use both pickups full on. The sound that works best in the band may not match your concept of a "great" bass tone so be prepared to have and use an open mind.

New strings will help immensely here so you have a baseline for comparison.
