I would go with envelope filter and compressor. If someone else would be asking I may change my answer depending on context.
I don't have a pre-amp with DI, but that would be the smart universal option for gigging.
I would add a chorus, but I can live without it.

I like to keep it simple and I don't use much effects on bass. My compressor is not sliders on, but when you need, you need it. The filter... It's way too fun, if you want to play funk or some shaged up stuff Les Claypool style it will always deliver
 
Comp and My DI that has a blendable drive. There, I broke the rules without actually breaking the rules...
Here’s where it gets real grey. Use an MS60B as your tuner (you said that didn’t count), you kinda have all that comes with it as a byproduct. So, there you go. Cake and you can eat it too enjoy!
